Azithromycin for injection – HPLC with ECD and UV

An updated application note has been released featuring the assay for ‘azithromycin for injection’. The method follows the USP 41 monograph using HPLC in combination with UV detection. It is now possible to run both the ECD and UV methods in the USP monographs for azithromycin formulations using the ALEXYS analyzer!

This is literally about the darkened side of grilled or roasted meat. About the chemistry that takes place with nitrate, nitrite, proteins and other amines forming nitrosamines. How well-prepared meat can have carcinogenic side effects, and why concentrations of nitrate and nitrite is of a concern.

Sulfur-containing antibiotics

A new application note has been released about the analysis of sulfamethoxazole and trimethoprim. The method is using HPLC with electrochemical detection. A multistep potential has been applied for stable and reproducible working conditions.

Carbohydrates – AOAC method

January 25, 2021

A new application note has been released for the analysis of carbohydrates in food, dietary supplements, pet food and animal feeds. We used the method as published by AOAC. The analysis is based on gradient separation on a High-Performance Anion-Exchange column followed by pulsed amperometric detection using a 4-step potential waveform.

Remote Assist Services

Recently we introduced a few new Remote Assisted Services. What we learned so far, is that the participating customers highly appreciate the remote sessions. For us this is an unexpected side effect, which encouraged us to develop a few more.

By doing the actual work, users are more involved in the session thus gaining a deeper understanding of the instruments.

SenCell or FlexCell?

Over the past year we have received questions what flow cell to use for the analysis of carbohydrates. Method of analysis is HPLC with Pulsed Amperometric Detection (HPAEC-PAD) using a flow cell with gold electrode and HyREF. Both FlexCell and SenCell will do, here is what we think how to choose:

FlexCell: our first choice for 3-step pulsed amperometric detection. It has a high pH inert HyREF reference electrode and an exchangeable working electrode. This is a particularly useful feature to cope with the normal wear of the gold electrode. Taking out the electrode to polish or replace is easy.

SenCell: is recommended in case a 4-step pulse can be used with minimum wear of the gold electrode. It also has an inert HyREF reference electrode, and in most applications it has a bit better signal-to-noise ratio. Therefore, in most recent application notes we now use the SenCell.

Histamine – neurotransmitter and fresh fish

Histamine is one of those substances with a multitude of functions in the body. Probably the most annoying is its capability to trigger an immune response followed by itching or hay fever and such. As a neurotransmitter it plays a role to control the sleep-wake cycle.

It also plays a role in a totally different field, in fish products. Histamine levels rise with insufficient cooling or poor hygiene. It says something about freshness and fish quality for consumption.

LNAAs – How Food controls the Brain

Did you know that behavior can be affected by the food we eat? In agriculture this has been known for quite some time. By adding certain amino acids to animal food it has been shown that animals have less stress and aggression.

Recently, we published a method for analysis of LNAAs – Large Neutral Amino Acids including  Tryptophan, Tyrosine, and Phenylalanine – that play a key role in this. Click to download.

New FlexCell meets USP/EP requirement

We have recently developed a new FlexCell with Stainless Steel Auxiliary electrode especially for USP and EP methods. It combines the flexibility of an exchangeable gold working electrode with the USP/EP requirement for stainless steel AUX. The maintenance free & pH inert HyREF works very well for aminoglycosides or other carbo’s that are analyzed under alkaline conditions.

What Chromeleon license is needed?

We have some background information about the Antec ECD driver for Chromeleon™ CDS from Thermo Fisher Scientific and the Chromeleon “Class 3” license requirement.

Currently, several flavors of Chromeleon are available. First there is a choice between Workstation (standalone PC) or Enterprise (multi-PC Network) license. Then there are Class 1, 2 and 3 licenses:

  • Class 1: all Thermo Fisher Scientific instruments
  • Class 2: third party GC’s
  • Class 3: third  party LC’s

A Class 3 license automatically covers all Class 1 devices as well. Check what version of Chromeleon is being used. In case of class 1, contact Thermo Fisher Scientific as it should be upgraded to Class 3 to be able to work with an Antec ECD.

Checklist for Antec users

We created two checklists for Antec users to keep track of the daily maintenance and checks in HPLC/ECD. Particularly useful in troubleshooting are the daily entries for key parameters such as I-cell, pressure reading and the system suitability result.

We have a checklist for ALEXYS Analyzers, and one for 3rd party HPLC with Antec ECD which has less detail on pump and autosampler.
