The ALEXYS Neurotransmitter Analyzer can be used for fast and sensitive detection of basal acetylcholine (ACh) levels in microdialysis samples. Analysis of ACh by HPLC-ECD is based on an ion-pairing separation, followed by post-column enzymatic conversion to hydrogen peroxide with acetylcholinesterase (AChE) and choline oxidase (ChO). Both enzymes are covalently bound to a stationary phase in an immobilized enzyme reactor (IMER). After conversion, the hydrogen peroxide is detected using the DECADE Elite electrochemical detector with Sencell on a Pt electrode. The method is optimized for analysis of small sample volumes at low nanomolar detection limit.
