Le regioni esterne della proteina ripiegata subiscono un facile scambio di idrogeno (H) con il deuterio (D). Nelle fasi successive del flusso di lavoro, le proteine vengono dispiegate e la sequenza aminoacidica viene delucidata mediante MS. Gli amminoacidi con deuterio provengono ovviamente dalla regione esterna della proteina ripiegata. Per ottenere risultati affidabili \u00e8 importante che la reazione di scambio H\/D venga interrotta (quenched) e che i cambiamenti vengano conservati, una volta che lo svolgimento delle proteine e le fasi successive hanno luogo.<\/p>\n<p>La riduzione elettrochimica controllata dei legami S-S \u00e8 stata applicata con successo per il dispiegamento di proteine pi\u00f9 grandi come gli anticorpi, sostituendo la dura riduzione chimica che spesso comprometteva lo scambio H\/D. L&#8217;uso di agenti riducenti classici come DTT o TCEP \u00e8 spesso inefficace in condizioni di quenching (pH acido e bassa temperatura). Pertanto, l&#8217;uso di ROXY EC con i suoi elettrodi proprietari a base di titanio per la scissione delicata e controllata del legame S-S apre nuove opportunit\u00e0 nei flussi di lavoro HDX\/MS, con conseguente aumento della copertura delle sequenze.<\/p>\n<p>Le due figure mostrano la copertura della sequenza per il Nerve Growth Factor-\u03b2 con riduzione chimica ed elettrochimica (EC). L&#8217;integrazione della cella di flusso elettrochimica nel flusso di lavoro HDX\/MS ha permesso di ottenere una copertura di sequenza molto pi\u00f9 elevata rispetto alla riduzione chimica (99% contro 46%), consentendo una caratterizzazione molto pi\u00f9 completa dell&#8217;NGF.
