Les r\u00e9gions ext\u00e9rieures de la prot\u00e9ine repli\u00e9e subissent un \u00e9change facile d&rsquo;hydrog\u00e8ne (H) par du deut\u00e9rium (D). Dans les \u00e9tapes suivantes du flux de travail, les prot\u00e9ines sont d\u00e9pli\u00e9es et la s\u00e9quence d&rsquo;acides amin\u00e9s est \u00e9lucid\u00e9e par SM. Les acides amin\u00e9s contenant du deut\u00e9rium proviennent \u00e9videmment de la r\u00e9gion ext\u00e9rieure de la prot\u00e9ine repli\u00e9e. Pour obtenir des r\u00e9sultats fiables, il est important que la r\u00e9action d&rsquo;\u00e9change H\/D soit arr\u00eat\u00e9e (quench\u00e9e), et que les changements soient pr\u00e9serv\u00e9s, une fois que le d\u00e9pliage des prot\u00e9ines et les \u00e9tapes suivantes ont lieu.<\/p>\n<p>La r\u00e9duction \u00e9lectrochimique contr\u00f4l\u00e9e des liaisons S-S a \u00e9t\u00e9 appliqu\u00e9e avec succ\u00e8s pour le d\u00e9pliage de prot\u00e9ines plus grandes telles que les anticorps, rempla\u00e7ant ainsi la r\u00e9duction chimique brutale qui compromettait souvent l&rsquo;\u00e9change H\/D. L&rsquo;utilisation d&rsquo;agents r\u00e9ducteurs classiques tels que le DTT ou le TCEP est souvent inefficace dans des conditions de quenching (pH acide et basse temp\u00e9rature). Par cons\u00e9quent, l&rsquo;utilisation de ROXY EC avec ses \u00e9lectrodes exclusives \u00e0 base de titane pour le clivage doux et contr\u00f4l\u00e9 de la liaison S-S ouvre de nouvelles possibilit\u00e9s dans les flux de travail HDX\/MS, ce qui permet d&rsquo;obtenir une couverture de s\u00e9quence nettement sup\u00e9rieure.<\/p>\n<p>Les deux figures montrent la couverture de la s\u00e9quence du Nerve Growth Factor-\u03b2 avec r\u00e9duction chimique et \u00e9lectrochimique (EC). L&rsquo;int\u00e9gration de la cellule \u00e0 flux \u00e9lectrochimique dans le flux de travail HDX\/MS a donn\u00e9 lieu \u00e0 une couverture de s\u00e9quence beaucoup plus \u00e9lev\u00e9e que la r\u00e9duction chimique (99 % contre 46 %), permettant une caract\u00e9risation beaucoup plus compl\u00e8te du NGF.
