La r\u00e9duction des anticorps en cha\u00eenes l\u00e9g\u00e8res (Lc) et en cha\u00eenes lourdes (Hc) est r\u00e9alis\u00e9e par la m\u00e9thode \u00e9lectrochimique pr\u00e9c\u00e9demment publi\u00e9e, bas\u00e9e sur des impulsions de potentiel \u00e0 onde carr\u00e9e appliqu\u00e9es \u00e0 l&rsquo;\u00e9lectrode de Ti exclusive d&rsquo;Antec. Ensuite, la cyst\u00e9ine libre g\u00e9n\u00e9r\u00e9e par la r\u00e9duction \u00e9lectrochimique de l&rsquo;anticorps peut \u00eatre conjugu\u00e9e. Enfin, la cyst\u00e9ine libre sur Lc et Hc peut \u00eatre tr\u00e8s rapidement et s\u00e9lectivement r\u00e9-oxyd\u00e9e pour reformer l&rsquo;anticorps avec un rendement \u00e9lev\u00e9. Les spectres MS ci-dessous montrent les produits de la r\u00e9action (Lc et Hc) apr\u00e8s la mise en marche de la cellule du r\u00e9acteur (B et D).<\/p>\n<p>Infusion d&rsquo;une solution d&rsquo;anticorps acidifi\u00e9e dans le spectrom\u00e8tre de masse avec le \u03bc-PrepCell \u00e9teint (A) a donn\u00e9 lieu \u00e0 une enveloppe de charges s&rsquo;\u00e9talant entre m\/z 2200 et 4000, l&rsquo;anticorps intact portant environ 40 \u00e0 60 charges. Quand la cellule a \u00e9t\u00e9 allum\u00e9e (B), un d\u00e9placement de l&rsquo;enveloppe de charge de l&rsquo;anticorps vers les m\/z inf\u00e9rieurs (~1000-2500) a \u00e9t\u00e9 observ\u00e9.<\/p>\n<p>Le spectre (C) r\u00e9sulte de la d\u00e9convolution du signal MS (A) obtenu avec la cellule OFF et confirme que l&rsquo;anticorps est intact. D a \u00e9t\u00e9 obtenu avec la cellule allum\u00e9e, et montre principalement des esp\u00e8ces avec des poids mol\u00e9culaires d&rsquo;environ 25 et 50 kDa correspondant aux masses de Lc et Hc. La tr\u00e8s faible intensit\u00e9 du signal MS pour les esp\u00e8ces dont le poids mol\u00e9culaire est sup\u00e9rieur \u00e0 50 kDa confirme \u00e9galement la r\u00e9duction s\u00e9lective des
