La r\u00e9duction des liaisons disulfure intra- et intermol\u00e9culaires est n\u00e9cessaire pour une caract\u00e9risation et une affectation r\u00e9ussies des sites de liaison par MS. La r\u00e9duction hors ligne est r\u00e9alis\u00e9e \u00e0 l&rsquo;aide d&rsquo;agents chimiques hautement concentr\u00e9s, par <em>exemple le<\/em> dithiothreitol (DTT), qui doivent \u00eatre \u00e9limin\u00e9s avant l&rsquo;analyse LC\/MS. On peut \u00e9galement utiliser des agents r\u00e9ducteurs sans thiol tels que le TCEP (tris (2-carboxy\u00e9thyl) phosphine). Cependant, la pr\u00e9paration des \u00e9chantillons reste laborieuse et difficile \u00e0 combiner avec la LC\/MS en ligne. En outre, la possibilit\u00e9 de r\u00e9duire les liaisons disulfure en ligne peut \u00eatre b\u00e9n\u00e9fique pour la d\u00e9termination des arrangements de liaisons disulfure ou la strat\u00e9gie de prot\u00e9omique descendante, qui repose sur la fragmentation de prot\u00e9ines intactes sans digestion enzymatique.<\/p>\n<p>Enfin, l&rsquo;utilisation (LC)\/EC\/MS montre un grand potentiel pour l&rsquo;affectation rapide des liaisons S-S dans les produits biopharmaceutiques. Les syst\u00e8mes ROXY EC et ROXY EC\/LC, avec leurs \u00e9lectrodes exclusives \u00e0 base de titane, sont les instruments id\u00e9aux pour r\u00e9aliser de telles \u00e9tudes.<\/p>\n<p><strong>R\u00e9duction des liaisons disulfure de l&rsquo;insuline assist\u00e9e par voie \u00e9lectrochimique<\/strong><\/p>\n<p>L&rsquo;insuline, une petite prot\u00e9ine de 5733 Da, est utilis\u00e9e comme compos\u00e9 mod\u00e8le pour la r\u00e9duction S-S. Il est constitu\u00e9 de 51 acides amin\u00e9s formant deux cha\u00eenes, A et B, et contient 3 liaisons disulfure.
