Le r\u00e9acteur est en fait une cellule \u00e9lectrochimique, un petit potentiel est appliqu\u00e9 sur les \u00e9lectrodes pour entra\u00eener la r\u00e9action. La sortie de la cellule est coupl\u00e9e \u00e0 un MS pour l&rsquo;analyse des produits.<\/p>\n<p>Plusieurs param\u00e8tres sont importants pour la r\u00e9ussite d&rsquo;une exp\u00e9rience EC\/MS. Le premier est le <strong>potentiel appliqu\u00e9<\/strong>, il fournit l&rsquo;\u00e9nergie n\u00e9cessaire pour qu&rsquo;une r\u00e9action se produise. Un potentiel positif pour les r\u00e9actions d&rsquo;oxydation et n\u00e9gatif pour les r\u00e9actions de r\u00e9duction. Une fois que les conditions sont \u00e9tablies et que la r\u00e9action se produit, augmenter encore le potentiel n&rsquo;acc\u00e9l\u00e9rera pas la r\u00e9action et ne g\u00e9n\u00e9rera pas plus de produit. La raison en est expliqu\u00e9e dans les lois traditionnelles de l&rsquo;\u00e9lectrochimie. Le facteur limitant n&rsquo;est pas le potentiel, c&rsquo;est la diffusion du substrat de la solution \u00e0 l&rsquo;\u00e9lectrode.<\/p>\n<p>Le deuxi\u00e8me param\u00e8tre important est la <strong>composition du solvant<\/strong>. La valeur du pH et le pourcentage en solvant organique peuvent avoir une grande influence. Pour la r\u00e9duction des prot\u00e9ines par exemple, l&rsquo;acide formique est utilis\u00e9 pour fixer le pH. Mais aussi parce qu&rsquo;il engendre un interm\u00e9diaire r\u00e9actif qui catalyse la r\u00e9action, d\u00e9crite plus en d\u00e9tail par <a href=\"https:\/\/link.springer.com\/article\/10.1007%2Fs00216-018-1447-2\">B. Stocks et J. Melanson (<span data-test=\"article-publication-year\">2019<\/span>)<\/a>.<\/p>\n<p>De plus, le <strong>d\u00e9bit<\/strong> est important, typiquement \u00e0 un d\u00e9bit plus faible, un pourcentage relativement plus important du substrat sera converti en produit. Les produits biopharmaceutiques sont des m\u00e9dicaments qui sont produits dans des syst\u00e8mes vivants gr\u00e2ce \u00e0 la biotechnologie. Il s&rsquo;agit de prot\u00e9ines complexes de poids mol\u00e9culaire \u00e9lev\u00e9, telles que les prot\u00e9ines de fusion, les prot\u00e9ines de bioaffinit\u00e9, les anticorps (mAbs) et les conjugu\u00e9s anticorps-m\u00e9dicaments (ADC).<\/p>\n<p>Avec le nombre croissant de produits biopharmaceutiques apparaissant sur le march\u00e9, il existe une demande croissante de strat\u00e9gies efficaces et automatis\u00e9es pour l&rsquo;\u00e9lucidation structural. Cela peut \u00eatre un v\u00e9ritable d\u00e9fi. Ce n&rsquo;est pas seulement la structure primaire, la cartographie des acides amin\u00e9s qui est importante. Elle concerne \u00e9galement la structure d&rsquo;ordre sup\u00e9rieur (HOS) d\u00e9finie par les ponts disulfures internes et les modifications post-traductionnelles telles que les phosphorylations, les ac\u00e9tylations, les glycosylations, etc. La structure est ce qui d\u00e9finit l&rsquo;activit\u00e9 biologique (revue : <a href=\"https:\/\/link.springer.com\/article\/10.1007\/s10337-019-03796-9\">D. Vir\u00e1g et al, 2020<\/a>).<\/p>\n<p>Les liaisons disulfures dans les prot\u00e9ines assurent la stabilit\u00e9 et le reploiement, et sont donc d&rsquo;une importance capitale pour la fonctionnalit\u00e9 biologique. Dans les anticorps monoclonaux, par exemple, les liaisons disulfure maintiennent ensemble la chaine lourde et la chaine l\u00e9g\u00e8re. Il est donc important de v\u00e9rifier la localisation et la connectivit\u00e9 des disulfures dans les prot\u00e9ines th\u00e9rapeutiques. Cela se fait g\u00e9n\u00e9ralement en r\u00e9duisant les liaisons disulfure et en analysant les changements dans les spectres MS (revue : <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC5857437\/\">J.
