Las regiones externas de la prote\u00edna plegada sufren un f\u00e1cil intercambio de Hidr\u00f3geno (H) por Deuterio (D). En los siguientes pasos del flujo de trabajo, las prote\u00ednas se despliegan y la secuencia de amino\u00e1cidos se dilucida mediante EM. Los amino\u00e1cidos con deuterio proceden obviamente de la regi\u00f3n externa de la prote\u00edna plegada. Para obtener resultados fiables, es importante que la reacci\u00f3n de intercambio H\/D se detenga (se apague) y se conserven los cambios, una vez que se produzca el desdoblamiento de las prote\u00ednas y los pasos siguientes.<\/p>\n<p>La reducci\u00f3n electroqu\u00edmica controlada de los enlaces S-S se ha aplicado con \u00e9xito para el desdoblamiento de prote\u00ednas de mayor tama\u00f1o, como los anticuerpos, sustituyendo a la dura reducci\u00f3n qu\u00edmica que a menudo pon\u00eda en peligro el intercambio H\/D. El uso de agentes reductores cl\u00e1sicos como el DTT o el TCEP suelen ser ineficaces en condiciones de enfriamiento (pH \u00e1cido y baja temperatura).
