genera cadenas ligeras y pesadas<\/li>\n<li>R\u00e1pido &#8211; segundos frente a horas con enfoques qu\u00edmicos (por ejemplo, TCEP, DTT)<\/li>\n<\/ul>\n<p>Este novedoso enfoque electroqu\u00edmico proporciona un m\u00e9todo r\u00e1pido y eficaz para la reducci\u00f3n selectiva, conjugaci\u00f3n y reoxidaci\u00f3n de anticuerpos sin necesidad de productos qu\u00edmicos reductores. La reducci\u00f3n de los anticuerpos en sus cadenas ligeras (Lc) y pesadas (Hc) se realiza por el m\u00e9todo electroqu\u00edmico publicado anteriormente, basado en pulsos de potencial de onda cuadrada aplicados al electrodo de Ti patentado por Antec. Posteriormente, se puede conjugar la ciste\u00edna libre generada por la reducci\u00f3n electroqu\u00edmica del anticuerpo. Por \u00faltimo, la ciste\u00edna libre tanto en Lc como en Hc puede reoxidarse muy r\u00e1pida y selectivamente para reformar el anticuerpo en altos rendimientos. Los siguientes espectros MS muestran los productos de reacci\u00f3n (Lc y Hc) tras encender la c\u00e9lula del reactor (B y D).<\/p>\n<p>Infusi\u00f3n de una soluci\u00f3n acidificada de anticuerpos en el espectr\u00f3metro de masas con el \u03bc-PrepCell apagado. (A) dio lugar a una envoltura de carga que se extend\u00eda entre m\/z 2200 y 4000, con el anticuerpo intacto portando aproximadamente de 40 a 60 cargas. Cuando la c\u00e9lula estaba encendida (B), se observ\u00f3 un desplazamiento de la envoltura de carga del anticuerpo hacia m\/z inferiores (~1000-2500).<\/p>\n<p>El espectro (C) resulta de la deconvoluci\u00f3n de la se\u00f1al MS (A) obtenida con la c\u00e9lula OFF y confirma que el anticuerpo est\u00e1 intacto. D se obtuvo con la c\u00e9lula encendida, y muestra principalmente especies con pesos moleculares de aproximadamente 25 y 50 kDa correspondientes a las masas de Lc y Hc. La muy baja intensidad de la se\u00f1al de EM para especies con pesos moleculares superiores a 50 kDa confirma adem\u00e1s la reducci\u00f3n selectiva de los enlaces disulfuro intercadena del
