Estabilizan la estructura tridimensional de la prote\u00edna y son cruciales para su funci\u00f3n biol\u00f3gica. La reducci\u00f3n de los enlaces disulfuro intra e intermoleculares es necesaria para caracterizar y asignar con \u00e9xito los sitios de enlace mediante EM. La reducci\u00f3n fuera de l\u00ednea se realiza utilizando agentes qu\u00edmicos altamente concentrados, por <em>ejemplo,<\/em> ditiotreitol (DTT), que deben eliminarse antes del an\u00e1lisis LC\/MS. Como alternativa, pueden utilizarse agentes reductores libres de tioles, como la TCEP (tris (2-carboxietil) fosfina). Sin embargo, la preparaci\u00f3n de las muestras sigue siendo laboriosa y dif\u00edcil de combinar con la LC\/MS en l\u00ednea. Adem\u00e1s, la posibilidad de reducir en l\u00ednea los enlaces disulfuro puede ser beneficiosa para la determinaci\u00f3n de las disposiciones de los enlaces disulfuro o la estrategia de prote\u00f3mica top down, que se basa en la fragmentaci\u00f3n de prote\u00ednas intactas sin digesti\u00f3n enzim\u00e1tica.<\/p>\n<p>Por \u00faltimo, el uso (LC)\/EC\/MS muestra un gran potencial para la asignaci\u00f3n r\u00e1pida de enlaces S-S en biof\u00e1rmacos. Los sistemas ROXY EC y ROXY EC\/LC, con sus electrodos patentados basados en titanio, son los instrumentos ideales para realizar este tipo de estudios.<\/p>\n<p><strong>Reducci\u00f3n electroqu\u00edmica asistida de los enlaces disulfuro en la insulina<\/strong><\/p>\n<p>La insulina, una peque\u00f1a prote\u00edna de 5733 Da, se utiliza como compuesto modelo para la reducci\u00f3n S-S. Consta de 51 amino\u00e1cidos que forman dos cadenas, A y B, y contiene 3 enlaces disulfuro.
