TCEP, DTT)<\/li>\n<\/ul>\n<p>Dieser neuartige elektrochemische Ansatz bietet eine schnelle und effiziente Methode f\u00fcr die selektive Reduktion, Konjugation und Reoxidation von Antik\u00f6rpern, ohne dass reduzierende Chemikalien ben\u00f6tigt werden. Die Reduktion von Antik\u00f6rpern in ihre leichten Ketten (Lc) und schweren Ketten (Hc) wird mit der bereits ver\u00f6ffentlichten elektrochemischen Methode durchgef\u00fchrt, die auf Rechteckpotentialimpulsen basiert, die an die von Antec entwickelte Ti-Elektrode angelegt werden. Anschlie\u00dfend kann das freie Cystein, das bei der elektrochemischen Reduktion des Antik\u00f6rpers entsteht, konjugiert werden. Schlie\u00dflich kann das freie Cystein sowohl auf Lc als auch auf Hc sehr schnell und selektiv reoxidiert werden, um den Antik\u00f6rper in hoher Ausbeute neu zu bilden. B. bi-spezifische Antik\u00f6rper oder Antik\u00f6rper-Wirkstoff-Konjugate. Die MS-Spektren unten zeigen die Reaktionsprodukte (Lc und Hc) nach dem Einschalten der Reaktorzelle (B und D).<\/p>\n<p>Infusion einer anges\u00e4uerten Antik\u00f6rperl\u00f6sung in das Massenspektrometer bei ausgeschalteter \u03bc-PrepCell (A) f\u00fchrte zu einer Ladungsh\u00fcllkurve, die sich zwischen m\/z 2200 und 4000 ausbreitet, wobei der intakte Antik\u00f6rper etwa 40 bis 60 Ladungen tr\u00e4gt. Wenn die Zelle eingeschaltet wurde (B) wurde eine Verschiebung der Ladungsh\u00fclle des Antik\u00f6rpers zu niedrigeren m\/z-Werten (~1000-2500) beobachtet.<\/p>\n<p>Das Spektrum (C) ergibt sich aus der Entfaltung des MS-Signals (A), das mit der ausgeschalteten Zelle erhalten wurde, und best\u00e4tigt, dass der Antik\u00f6rper intakt ist. D wurde bei eingeschalteter Zelle erhalten und zeigt haupts\u00e4chlich Spezies mit Molekulargewichten von etwa 25 und 50 kDa, die den Massen von Lc und Hc entsprechen. Die sehr geringe Intensit\u00e4t des MS-Signals f\u00fcr Spezies mit einem Molekulargewicht von mehr als 50 kDa best\u00e4tigt die selektive Reduktion der
