Die Reduktion von intra- und intermolekularen Disulfidbindungen ist ein kleiner, aber entscheidender Schritt im Arbeitsablauf f\u00fcr eine erfolgreiche Strukturaufkl\u00e4rung und Zuordnung der Bindungsstellen durch MS. Die Methode basiert auf einem elektrischen Potential zur Ausl\u00f6sung der Reduktionsreaktion, es werden keine reduzierenden Chemikalien verwendet.<\/p>\n<p>Traditionell wird die Reduktion mit konzentrierten Chemikalien durchgef\u00fchrt, z <em>.<\/em> B. mit Dithiothreitol (DTT), das vor der LC\/MS-Analyse entfernt werden muss. Alternativ k\u00f6nnen auch thiolfreie Reduktionsmittel wie TCEP (Tris(2-carboxyethyl)phosphin) verwendet werden. Die Probenvorbereitung ist jedoch nach wie vor aufw\u00e4ndig und l\u00e4sst sich nur schwer mit der Online-LC\/MS kombinieren.
